Tissue specificity of Drosophila P element transposition is regulated at the level of mRNA splicing.

چکیده
We show that the germline specificity of P element transposition is controlled at the level of mRNA splicing and not at the level of transcription. In the major P element RNA transcript, isolated from somatic cells, the first three open reading frames are joined by the removal of two introns. Using in vitro mutagenesis and genetic analysis we demonstrate the existence of a third intron whose removal is required for transposase production. We propose that this intron is only removed in the germline and that its removal is the sole basis for the germline restriction of P element transposition.

The basis for germline specificity of the hobo transposable element in Drosophila melanogaster.
Previous results suggested that the hobo transposable element is active predominantly in the germline of Drosophila. We investigate germline restriction of hobo transposition by testing in vitro modified elements for their ability to mobilize marked elements in vivo. Although intact hobo elements are germline specific, an hsp70 promoter-hobo transposase fusion is active in the soma. Analysis of...

Alternative splicing of pre-mRNA is a highly regulated process that allows cells to change their genetic informational output. These changes are mediated by protein factors that directly bind specific pre-mRNA sequences. Although much is known about how these splicing factors regulate pre-mRNA splicing events, comparatively little is known about the regulation of the splicing factors themselves...
متن کاملTiming and targeting of P-element local transposition in the male germline cells of Drosophila melanogaster.
The P element in Drosophila melanogaster preferentially transposes into nearby sites. The local insertions display a preferential orientation toward the starting element. We investigated the mechanism of the P-element local transposition by isolating and characterizing local insertions in the male germline. We designed a genetic screen employing a marker gene that is carried in the P element an...
متن کاملAlternative splicing of the Drosophila Dscam pre-mRNA is both temporally and spatially regulated.
The Drosophila melanogaster Down syndrome cell adhesion molecule (Dscam) gene encodes an axon guidance receptor that can express 38,016 different mRNAs by virtue of alternative splicing. The Dscam gene contains 95 alternative exons that are organized into four clusters of 12, 48, 33, and 2 exons each. Although numerous Dscam mRNA isoforms can be synthesized, it remains to be determined whether ...
متن کاملRegulation of tissue-specific P-element pre-mRNA splicing requires the RNA-binding protein PSI.
Binding of a multiprotein complex to a 5' exon inhibitory element appears to repress splicing of the Drosophila P-element third intron (IVS3) in the soma. We have purified 97- and 50-kD proteins that interact specifically with the inhibitory element using RNA affinity chromatography. Antibodies specific for the 97-kD protein relieve inhibition of IVS3 splicing in somatic extracts, providing dir...
